Keep three quantities separate
Milligrams (mg) and micrograms (mcg) measure mass: 1 mg = 1,000 mcg. Milliliters (mL) measure liquid volume. A U-100 syringe marking represents volume on that specific scale; it does not describe peptide mass or biological potency.
Concentration (mg/mL) = vial content (mg) ÷ final solution volume (mL)
Volume (mL) = target dose (mg) ÷ concentration (mg/mL)
U-100 marking = volume (mL) × 100
Use matching mass units before dividing. Convert mcg to mg by dividing by 1,000, or use mcg with a concentration expressed in mcg/mL. A vial labeled only in mg gives total content, not concentration. The FDA’s strength-conversion reference distinguishes amount per vial from amount per liquid volume.
A worked example
This is hypothetical arithmetic, not a suggested dose or preparation:
- A single-ingredient vial containing 10 mg in a confirmed final volume of 2 mL has a concentration of 5 mg/mL.
- An entered target dose of 0.25 mg equals 250 mcg.
- 0.25 mg ÷ 5 mg/mL = 0.05 mL, corresponding to mark 5 on U-100.
A matching scale alone does not establish measurability. Check the syringe’s capacity and smallest marking interval. A result between lines must not be rounded into a different amount. The U-100 units-to-mL calculator explains the volume mapping.
Split a full vial into equal injections
Instead of entering a target dose, choose a positive whole number of injections. The calculation divides the full vial equally:
Amount per injection (mg) = vial content (mg) ÷ injection count
Volume per injection (mL) = final mixed volume (mL) ÷ injection count
For example, dividing the same 10 mg vial in 2 mL into 20 equal injections gives 0.5 mg in 0.1 mL per injection, or 10 U-100 units. This is another hypothetical calculation, not a dose recommendation.
When you enter a target dose, the theoretical full-injection count is the whole-number part of vial content divided by target dose. Leftover peptide equals vial content minus the amount in those full injections. These figures assume a full vial and exclude liquid left in the vial or syringe.
Compare water amounts
The mixing calculator starts with your target dose or equal injection count, then vial content and syringe line spacing. It compares water amounts and estimated draws, with an “Easy to measure” preference among the measurable options. Select an option or enter your own water amount. The preference concerns measurement, not product compatibility or a medically appropriate mixture.
Estimated concentration (mg/mL) = vial content (mg) ÷ selected water (mL)
Estimated draw (mL) = target dose (mg) ÷ estimated concentration (mg/mL)
Or: estimated draw (mL) = selected water (mL) ÷ injection count
These formulas assume water amount equals final volume. The mixing syringe defaults to 3 mL capacity; that is neither a vial capacity nor a selected water amount. Both syringes require their actual line spacing. An optional “Maximum water for this vial” from product directions limits the comparison; leaving it blank does not establish a vial limit.
Plan a mix around a syringe draw
Use “Choose a syringe draw instead” to enter the U-100 line you want for each injection. Together with vial content and dose or injection count, this determines the final mixed volume:
Chosen draw (mL) = chosen U-100 units ÷ 100
Final mixed volume (mL) = vial content (mg) ÷ target dose (mg) × chosen draw (mL)
Or: final mixed volume (mL) = injection count × chosen draw (mL)
With the example above, 20 injections at a chosen 10 U-100 units (0.1 mL) each require a total mixed volume of 2 mL. Entering 10 mg, a target dose of 0.5 mg, and the same draw gives the same result.
Mass fields default to mg, and the syringe defaults to U-100, 1 mL (100 units). These are visible starting settings, not dose or mixing recommendations. Capacity and line spacing are editable. Line spacing starts at 1 unit for the insulin syringe and 0.1 mL for the 3 mL mixing syringe; check these settings against your syringe. Arithmetic and line checks use exact fractions internally; approximate decimal displays are identified.
Final volume is not automatically water to add
The concentration formula uses the total volume of the finished solution. The water comparison uses the selected water amount as an estimate of that volume. In the already-mixed calculator, “How much bacteriostatic water did you add?” provides the same kind of estimate. The estimated concentration is vial content divided by water added; the estimated draw follows from that concentration. For equal injections, the estimated volume per injection is water added divided by injection count. When a labeled concentration is available, enter that directly instead.
Added liquid and final volume need not match. See the methodology and sources for this distinction. The calculator does not apply a universal powder-volume adjustment or quantify the accuracy of a water-based estimate.
The calculated mixed volume cannot select a diluent, establish compatibility, or become preparation instructions. Use the specific product instructions to determine the appropriate liquid and amount to add, and whether the total volume fits the vial; ask a pharmacist if those instructions are unclear.
